Autoclave validation guidelines: IQ, OQ, PQ and routine testing

Updated 14 October 2026 · 3 min read

Autoclave validation guidelines answer one question: can you prove that your steam sterilizer, with your loads and your packaging, reliably delivers sterility? Validation is a documented programme of installation, operational and performance qualification, followed by routine monitoring and periodic revalidation. This article explains what autoclave validation is, what the process includes and what a hospital CSSD should document.

What is autoclave validation?

Sterility cannot be confirmed by testing the finished instrument, because testing would destroy the sterile barrier. Instead, the process is validated: evidence is gathered that the sterilizer, cycle and load configuration consistently achieve the required conditions, and then the process is controlled and monitored to stay within them.

The international reference for moist-heat sterilization of health care products is ISO 17665. European standards for steam sterilizers (EN 285 for large sterilizers, EN 13060 for small ones) define machine performance requirements. Your equipment supplier and local authority will indicate which documents apply; confirm current requirements with your regulator.

The validation process: IQ, OQ and PQ

Installation qualification (IQ) shows that the sterilizer was delivered and installed as specified: correct utilities, steam quality, drains, calibrated sensors, documentation and safety devices. Operational qualification (OQ) shows that the machine operates within its specified limits across its cycles, including air removal, leak rate, temperature distribution in the empty chamber and alarm functions.

Performance qualification (PQ) tests the actual loads: the heaviest and most difficult configurations of instrument sets, containers or wrapped packs, with temperature and pressure probes and biological indicators placed in the hardest-to-reach places. A written protocol states acceptance criteria before testing, and a report records the results and approval.

Autoclave validation protocol and checklist

Container loads need special attention. Rigid containers have more mass and different drying behaviour than wrapped trays, so they should be part of the PQ load set, using the container manufacturer's IFU (REDA sterilization containers). Parameters such as holding times are covered in steam sterilization temperature and time.

  • Scope, equipment identification, cycle definitions and responsibilities.
  • Calibration status of probes and gauges used in testing.
  • Steam quality, air removal and leak test results.
  • Load configurations, including worst-case sets and containers.
  • Temperature, pressure and holding time records for each cycle.
  • Biological and chemical indicator results.
  • Drying results: no visible moisture on packs or containers.
  • Deviations, corrective actions and final approval signatures.

Routine tests and autoclave validation frequency

Between validations, daily and per-load checks keep the process in control: a Bowie-Dick or equivalent air-removal test for pre-vacuum machines at the start of each day, review of each cycle's printout, chemical indicators in every pack and biological indicators at a frequency set by your policy. See the Bowie-Dick test for how the daily test works.

Revalidation is normally performed at least annually and after major repairs, relocation, software changes or a change in load type or packaging. Your policy and regulator define the exact frequency, so treat these as typical practice rather than a rule.

Records, responsibilities and next steps

Keep validation reports, calibration certificates, routine test results, maintenance logs and cycle records for the period required by local rules, and link every released pack to its load, as described in instrument tracking systems. Assign clear roles: a competent person to authorise validation, engineers to maintain machines, and CSSD supervisors to run routine tests.

Frequently asked questions

What is autoclave validation?

A documented programme proving that a sterilizer, cycle and load configuration consistently deliver the conditions needed for sterility, followed by routine monitoring.

What do IQ, OQ and PQ mean?

Installation, operational and performance qualification: correct installation, correct machine operation within limits, and proven results on real worst-case loads.

How often should an autoclave be revalidated?

Typically at least annually and after major repair, relocation or change of load or packaging, according to your policy and regulator.

Does validation replace daily tests?

No. Daily air-removal tests, cycle review and indicators remain essential between validations.
